功能与作用机制
FGF-4是一种有效的促有丝分裂原和形态发生素。其作用具有情境依赖性,因细胞类型、浓度以及其他因子的存在而异。
1、维持多能性与启动分化: 在胚胎干细胞和诱导多能干细胞中,FGF-4有助于维持多能性,并启动细胞向特定谱系(特别是中胚层谱系)分化。
2、肢芽发育: 这是其在胚胎发育中的经典作用。FGF-4在肢芽的顶端外胚层嵴中表达,通过刺激下方间充质细胞的增殖,对肢体的生长和模式形成至关重要。
3、中胚层分化: 驱动多能细胞向中胚层祖细胞分化,这些祖细胞可进一步分化为心肌细胞、肌肉、骨骼和结缔组织。
4、细胞存活与增殖: 促进多种细胞类型(包括成纤维细胞、内皮细胞和成肌细胞)的生长和存活。
